== Positive-incentive perspective == The positive-incentive perspective is an umbrella term for a set of theories presented as an alternative to the set-point theories of hunger and eating. The central assertion to the positive-incentive perspective is the idea that humans and other animals are not normally motivated to eat by energy deficits, but are instead motivated to eat by the anticipated pleasure of eating, or the positive-incentive value. According to this perspective, eating is controlled in much the same way as sexual behavior. Humans engage in sexual behavior, not because of an internal deficit, but instead because they have evolved to crave it. Similarly, the evolutionary pressures of unexpected food shortages have shaped humans and all other warm blooded animals to take advantage of food when it is present. It is the presence of good food, or the mere anticipation of it that makes one hungry.
=== Pyrolysis gas chromatography === Pyrolysis–gas chromatography–mass spectrometry is a method of chemical analysis in which the sample is heated to decomposition to produce smaller molecules that are separated by gas chromatography and detected using mass spectrometry. Pyrolysis is the thermal decomposition of materials in an inert atmosphere or a vacuum. The sample is put into direct contact with a platinum wire, or placed in a quartz sample tube, and rapidly heated to 600–1000 °C. Depending on the application even higher temperatures are used. Three different heating techniques are used in actual pyrolyzers: Isothermal furnace, inductive heating (Curie point filament), and resistive heating using platinum filaments. Large molecules cleave at their weakest points and produce smaller, more volatile fragments. These fragments can be separated by gas chromatography. Pyrolysis GC chromatograms are typically complex because a wide range of different decomposition products is formed. The data can either be used as fingerprints to prove material identity or the GC/MS data is used to identify individual fragments to obtain structural information. To increase the volatility of polar fragments, various methylating reagents can be added to a sample before pyrolysis. Besides the usage of dedicated pyrolyzers, pyrolysis GC of solid and liquid samples can be performed directly inside Programmable Temperature Vaporizer (PTV) injectors that provide quick heating (up to 30 °C/s) and high maximum temperatures of 600–650 °C. This is sufficient for some pyrolysis applications.
The second phase uses a modified MaxSub algorithm: a single 7 reside aligned pair in each proteins is used to orient the two full length protein structures to maximally superimpose these just these 7 C-alpha, then in this orientation it scans for any additional aligned pairs that are close in 3D. It re-orients the structures to superimpose this expanded set and iterates until no more pairs coincide in 3D. This process is restarted for every 7 residue window in the seed alignment. The output is the maximal number of atoms found from any of these initial seeds. This statistic is converted to a calibrated E-value for the similarity of the proteins. Mammoth makes no attempt to re-iterate the initial alignment or extend the high quality sub-subset. Therefore, the seed alignment it displays can't be fairly compared to DALI or TM align as it was formed simply as a heuristic to prune the search space. (It can be used if one wants an alignment based solely on local structure-motif similarity agnostic of long range rigid body atomic alignment.) Because of that same parsimony, it is well over ten times faster than DALI, CE and TM-align. It is often used in conjunction with these slower tools to pre-screen large data bases to extract the just the best E-value related structures for more exhaustive superposition or expensive calculations.

=== Wild-type GFP (wtGFP) === In the 1960s and 1970s, GFP, along with the separate luminescent protein aequorin (an enzyme that catalyzes the breakdown of luciferin, releasing light), was first purified from the jellyfish Aequorea victoria and its properties studied by Osamu Shimomura. In A. victoria, GFP fluorescence occurs when aequorin interacts with Ca2+ ions, inducing a blue glow. Some of this luminescent energy is transferred to the GFP, shifting the overall color towards green. However, its utility as a tool for molecular biologists did not begin to be realized until 1992 when Douglas Prasher reported the cloning and nucleotide sequence of wtGFP in Gene. The funding for this project had run out, so Prasher sent cDNA samples to several labs. The lab of Martin Chalfie expressed the coding sequence of wtGFP, with the first few amino acids deleted, in heterologous cells of E. coli and C. elegans, publishing the results in Science in 1994. Frederick Tsuji's lab independently reported the expression of the recombinant protein one month later. Remarkably, the GFP molecule folded and was fluorescent at room temperature, without the need for exogenous cofactors specific to the jellyfish. Although this near-wtGFP was fluorescent, it had several drawbacks, including dual peaked excitation spectra, pH sensitivity, chloride sensitivity, poor fluorescence quantum yield, poor photostability and poor folding at 37 °C (99 °F). == Extraction == Extraction using superheated water tends to be fast because diffusion rates increase with temperature. Organic materials tend to increase in solubility with temperature, but not all at the same rate. For example, in extraction of essential oils from rosemary and coriander, the more valuable oxygenated terpenes were extracted much faster than the hydrocarbons. Therefore, extraction with superheated water can be both selective and rapid, and has been used to fractionate diesel and woodsmoke particulates. Superheated water is being used commercially to extract starch material from marsh mallow root for skincare applications and to remove low levels of metals from a high-temperature resistant polymer. For analytical purposes, superheated water can replace organic solvents in many applications, for example extraction of PAHs from soils and can also be used on a large scale to remediate contaminated soils, by either extraction alone or extraction linked to supercritical or wet oxidation.
=== 2011 Sinaloa massacre === On 23 November 2011, a total of 26 bodies—16 of them burned to death—were located in several abandoned vehicles in Sinaloa. The incident began at early hours of the morning in Culiacán, Sinaloa with the discovery of a vehicle on fire. When the police forces managed to put down the flames, they found inside the vehicle a dozen bodies burned to death, and with wood remains on top of them. All of the victims were handcuffed. Later, at 07:00, anonymous calls from civilians notified the police that another vehicle in the northern city limits of Culiacán was on fire. The vehicle on fire was a Ford Ranger, and inside were four bodies with bulletproof vests and handcuffed. During the night, 10 more bodies were found throughout several municipalities. The killings were allegedly carried out by Los Zetas as a response to the massacres done by the Matazetas (CJNG) in Veracruz. Stratfor believes that this major move by Los Zetas into the territory of the Sinaloa Cartel demonstrates the Zeta's ability to attack the "heart of those cartels' territories."
== Research history == In the early 1900s, chemical messengers were crudely extracted from whole animal brains and tissues and studied for their physiological effects. In 1931, von Euler and Gaddum, used a similar method to try and isolate acetylcholine but instead discovered a peptide substance that induced physiological changes including muscle contractions and depressed blood pressure. These effects were not abolished using atropine, ruling out the substance as acetylcholine. In insects, proctolin was the first neuropeptide to be isolated and sequenced. In 1975, Alvin Starratt and Brian Brown extracted the peptide from hindgut muscles of the cockroach and found that its application enhanced muscle contractions. While Starratt and Brown initially thought of proctolin as an excitatory neurotransmitter, proctolin was later confirmed as a neuromodulatory peptide. David de Wied first used the term "neuropeptide" in the 1970s to delineate peptides derived from the nervous system.

Perfusion-rate-limited kinetics apply when the tissue membranes present no barrier to diffusion. === Validity === SPINA-GBeta significantly correlates with the M value in glucose clamp studies and (better than HOMA-Beta) with the two-hour value in oral glucose tolerance testing (OGTT), glucose rise in OGTT, subscapular skinfold, truncal fat content and the HbA1c fraction. It has the additional advantage that it circumvents the HOMA-blind zone, which renders the calculation of HOMA-Beta impossible if the fasting glucose concentration is 3.5 mmol/L (63 mg/dL) or below. Unlike HOMA-Beta, SPINA-Beta can be sensibly calculated in the whole range of measurements.

This butterfly is host specific for Japanese honeysuckle, but it may incidentally feed on other closely related plants, including Himalayan honeysuckle Leycesteria formosa. Oberea shirahatai is a Japanese honeysuckle host-specific beetle that feeds on the stems and leaves of Japanese honeysuckle. It was released in New Zealand in 2018 as another L. japonica-specialist biological control.
